Project title: Understanding the impact of inflammatory processes on ovarian ageing and fertility.
Aims: Ovarian ageing is defined by a progressive decline in oocyte/follicle number and quality, leading to loss of fertility and ovarian function, cycle irregularity and eventually menopause. The factors that contribute to the depletion of follicles throughout reproductive life are poorly characterised. However, with more women delaying childbearing, there is a growing need to elucidate the mechanisms underlying the reproductive ageing process. Several recent landmark studies have shown that chronic inflammation is associated with normal and pathological ageing in many organ systems, including the kidney, liver and gut. Furthermore, preliminary studies in my lab show that the supply of primordial follicles is dramatically and prematurely depleted in mouse models of chronic progressive low grade inflammation and generalised premature ageing (nfkb1-/-), whereas ovarian ageing is significantly delayed in mice in which inflammation is suppressed (asc-/-, nlrp3-/-). Therefore, the overall aim of my PhD project is to determine if inflammatory processes contribute to ovarian ageing.
